Background | 細(xì)胞凋亡早期改變發(fā)生在細(xì)胞膜表面,這些細(xì)胞膜表面的改變之一是磷脂酰絲氨酸(PS)從細(xì)胞膜內(nèi)轉(zhuǎn)移到細(xì)胞膜外,使PS暴露在細(xì)胞膜外表面。PS是一種帶負(fù)電荷的磷脂,正常主要存在于細(xì)胞膜的內(nèi)面,在細(xì)胞發(fā)生凋亡時細(xì)胞膜上的這種磷脂分布的不對稱性被破壞而使PS暴露在細(xì)胞膜外。Annexin V具有易于結(jié)合到磷脂類如PS的特性,對PS有高度的親和性。因此,該蛋白可充當(dāng)一敏感的探針檢測暴露在細(xì)胞膜表面的PS。PS轉(zhuǎn)移到細(xì)胞膜外不是凋亡所獨(dú)特的,也可發(fā)生在細(xì)胞壞死中。兩種細(xì)胞死亡方式間的差別是在凋亡的初始階段細(xì)胞膜是完好的,而細(xì)胞壞死在其早期階段細(xì)胞膜的完整性就破壞了。 |
